Prof. Li Qiang graduated from the Department of Chemical Engineering (DCE) of Tsinghua University, got his Master's degree in 1988 and went on with research and teaching in Tsinghua University since then. As a visiting scholar, he had been to the University of Tokyo and focused on gene recombination study in plant cells in the Life Science and Technology Department for a period of two years from 1997. Prof. Li had been the deputy head of the Medicine Institute of Tsinghua University, the deputy head of the Division of Biochemical Engineering and he is a syndic of the National Biological Process Simulation and Control Academy. Now he is the associate professor and doctoral supervisor of DCE as well as the director of the Laboratory of Antibody and Biocatalysis Engineering.
He has been engaged in the following research fields for about 30 years: Optimization of fermentation art in industrial microorganisms; Fermentation kinetics and new-style fermentation attachment; Kinetics of industrial enzyme catalysis and new methods of immobilization; Computer control in optimizing fermentation processes of enzymes and microorganisms; Expression of recombinant antigen, immunization, technology of hybridoma and animal cell culture; Construction of genetic engineering bacteria and genetic modification of strains based on metabolic engineering.
Prof. Li has published more than 70 papers in the field of Biochemical Engineering. He had been in charge of many important research projects, including one from the Natural Science Funds, four National Technological Key Projects and many projects of horizontal cooperation. The project "production of 7-ACA by two step enzymatic catalysis" has passed the expert review in 2007 and the novel technique has reached international advanced level, filling the gaps lacked in our country in this field. The technology for the enzymatic production of D-p-hydroxyphenylglycine by "one strain within two enzymes" method has been utilized in 125 cubic-meter-scale industrial reactors, which has doubled the production capacity without adding more equipment, and reached international advanced level. In recent years, Prof. Li has been engaged in the research of genetic engineering, including construction of recombinant strains for expressing industrial enzymes with high activity、metabolic engineering of industrial wild-type strains、construction of the universal expression system platform for synthetic biology research，and biological evaluation of biocompatible materials.
Research in Progress：
Engineering of biological reaction (required graduate course, 32 hours)
Basis of biochemical Engineering (required undergraduate course, 32 hours)
Engineering of cells cultivation (selective undergraduate course, 32 hours)
Industrial biocatalysis and synthetic biology：
Mainly include: construction of genetic recombinant strains，metabolic engineering of industrial strains，kinetics of industrial enzyme catalysis and new methods of purification and immobilization, kinetics of microbial fermentation, metabolic regulation of fermentation processes and optimization of technique，and amplification and industrialization of industrial biocatalysis. One main research interest is focused on construction of genetic recombinant strains for high efficient expression of industrial enzymes, as well as optimization of fermentation process and development of high-efficient medium for enzyme purification and new methods for enzyme immobilization, in order to reduce the production cost of industrial enzymes and promote the industrialization of biocatalysis. Another main research interest is focused on constructing universal expression elements for synthetic biology, testing the function of these expression elements in model hosts(E.coli, Bacillus subtilis, Bacillus megaterium, etc.) and industrial wild-type hosts, using these elements to do metabolic engineering and developing new strategy and technology for producing typical pharmaceutical products.
Immunization, Hybridoma Cell Technology and antibody detection：
Mainly includes: 1. Immunization method investigation, such as antigen immunization, polypeptide immunization, gene immunization; 2. Development of monoclonal antibodies for clinical diagnosis and polyclonal antibodies for detection of special proteins, using in vivo or large-scale in vitro culture of hybridoma cell lines obtained by the technology of immunization and hybridoma, as well as separation and purification technology.
Achievements and awards：
As the principal, Prof. Li completed the key projects of the national five year plan "New technology and devices for baker yeast production"(project No.85-08-04-04 ) and "New technology and process for Vitamin C production"(project No.85-08-04-01). For the first time in China Prof. Li put forward the strategy to link CO2 emission rate with carbohydrate metabolic rate, and realized the computer optimization, monitoring and control of fermentation in the scale of laboratory and 100m3 industrial production, by using fermentation kinetics model, monitoring model and control model. These projects were successfully completed and passed the examination.
For industrial biocatalysis, Prof. Li put forward the strategy to control the expression rate of heterologous proteins in construction of genetic recombinant strains, and successfully constructed the recombinant strain for producing 7-ACA by two step enzymatic catalysis, realizing the high-efficient and low-cost production of industrial enzymes. Further, the two-step enzymatic production technology was utilized in industry for the first time, realizing the total domestic production process of 7-ACA including the production, extraction and immobilization of enzymes and the biocatalytic production of 7-ACA. The total molar yield reached above 92%, which made the production cost lower than the chemical method and solved the pollution problem. This was the first domestic technology for industrial production of 7-ACA by two-step enzymatic biocatalysis, and passed the technique appraisement in January 2007.
Another achievement "Production technology for producing D-p-hydroxyphenylglycine by 'one strain within two enzymes' method" has been utilized in 125 cubic-meter-scale industrial reactors, which has doubled the production capacity without adding more equipment. The technique appraisement for this technology is in progress.
